FZD6-HER2 CAR T cells are an experimental cell therapy engineered to treat glioblastoma. These autologous or allogeneic T cells are retrovirally transduced to co-express a chimeric antigen receptor (CAR) targeting HER2 and the Wnt receptor Frizzled-6 (FZD6). Glioblastoma tumors often create a Wnt-rich microenvironment to support their own growth; by overexpressing FZD6, these CAR T cells are designed to hijack these tumor-derived Wnt ligands to promote their own survival, persistence, and memory maintenance. Preclinical studies indicate that FZD6 overexpression reduces T cell exhaustion and enhances cytotoxicity and intratumoral expansion compared to conventional HER2 CAR T cells. This strategy aims to exploit tumor-derived developmental cues to improve the durability of CAR T cell therapy in the immunosuppressive solid tumor microenvironment.
