GADLEN CD20 is a preclinical-stage bispecific gamma delta (γδ) T cell engager developed by Shattuck Labs for the treatment of antibody-mediated autoimmune diseases and B-cell malignancies. The compound is a bi-functional fusion protein comprising heterodimeric BTN2A1 and BTN3A1 extracellular domains fused via an inert Fc linker to a single-chain variable fragment (scFv) targeting the CD20 antigen. Its mechanism of action involves the selective recruitment and activation of Vγ9Vδ2 T cells to induce the lysis of CD20-positive B cells. Preclinical studies in humanized mice and non-human primates have demonstrated rapid and extensive B cell depletion (greater than 99%) with a favorable safety profile and no evidence of cytokine release syndrome.
