Overview

Galiximab + rituximab is a combination of two monoclonal antibodies investigated primarily for the treatment of follicular non-Hodgkin lymphoma. Galiximab is a primatized IgG1 lambda monoclonal antibody targeting CD80, a costimulatory molecule on B cells and some lymphomas. It blocks CD80–CD28 interactions, induces antibody-dependent cellular cytotoxicity (ADCC), inhibits cellular proliferation, and upregulates apoptotic proteins[1][2]. Rituximab is an anti-CD20 chimeric monoclonal antibody that depletes B cells by binding to CD20 and mediating ADCC, complement-dependent cytotoxicity (CDC), and apoptosis[5][6]. The combination has shown higher response rates than either agent alone in relapsed or treatment-naive follicular lymphoma patients[2][3][4]. Clinical trials demonstrated that the regimen was generally well tolerated with manageable infusion-related adverse events; however, phase 3 development for this indication was terminated[7].
