GaRD

GaRD is a combination drug consisting of Gallium Nitrate, Rituximab, and Dexamethasone. It was investigated as an experimental treatment for relapsed or refractory Diffuse Large B-cell Lymphoma. Rituximab is a chimeric monoclonal antibody that targets the CD20 antigen on B cells, leading to cell death through mechanisms such as complement-dependent cytotoxicity, antibody-dependent cell-mediated cytotoxicity, and direct apoptosis. Dexamethasone is a synthetic glucocorticoid that exerts anti-inflammatory and immunosuppressive effects by binding to intracellular glucocorticoid receptors and modulating gene expression. Gallium Nitrate acts as an iron mimetic, disrupting cellular iron homeostasis, inhibiting ribonucleotide reductase and protein tyrosine phosphatase, and inducing apoptosis in cancer cells.
