Gecacitinib hydrochloride (SHR0302) is a potent and selective small-molecule inhibitor of Janus kinase 1 (JAK1). In this therapeutic combination, it is paired with peginterferon alfa-2b, a pegylated cytokine, for the treatment of polycythemia vera (PV). Gecacitinib works by modulating the JAK-STAT signaling pathway, which is a key driver in the pathogenesis of myeloproliferative neoplasms. Peginterferon alfa-2b exerts antiproliferative effects on the malignant hematopoietic clone and stimulates the immune system. This combination is currently being evaluated in clinical trials (such as NCT06344104) to determine if the dual inhibition of JAK signaling and cytokine-mediated growth control provides superior hematological and molecular responses in PV patients compared to standard therapies.
