Gicasitinib (also known as ZG19018) is an orally bioavailable small molecule Janus kinase (JAK) inhibitor developed by Suzhou Zelgen Biopharmaceutical. It specifically targets the JAK1 isoform, which plays a critical role in the signaling pathways of various pro-inflammatory cytokines involved in the pathogenesis of autoimmune and inflammatory diseases. By inhibiting JAK1, gicasitinib modulates the JAK-STAT signaling cascade, thereby reducing the immune-mediated inflammation characteristic of conditions such as alopecia areata and atopic dermatitis.
