Glofitamab is a bispecific monoclonal antibody used for the treatment of adults with relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and large B-cell lymphoma (LBCL), including cases arising from follicular lymphoma, after at least two prior lines of therapy[1][3][5]. Marketed under the brand name Columvi, it was developed by Roche and is manufactured by Genentech. Glofitamab works as a bispecific T-cell engager antibody that simultaneously binds to CD20 antigen on B cells and CD3 antigen on T cells. This dual binding brings T cells into close proximity with malignant B cells, leading to T cell activation, proliferation, and targeted lysis of cancerous B cells[7][8]. The drug has shown significant efficacy in clinical trials for patients who have not responded to other treatments. It carries a boxed warning for cytokine release syndrome due to its immune-activating mechanism[2][5].
