Overview

Glofitamab + obinutuzumab is a combination regimen used primarily for the treatment of adult patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL) after two or more lines of systemic therapy. Glofitamab is a bispecific monoclonal antibody that binds to both CD20 on B cells and CD3 on T cells, bringing T cells into close proximity with malignant B cells to promote their destruction. Obinutuzumab is a humanized monoclonal antibody targeting CD20, leading to direct cell death and immune-mediated cytotoxicity against B lymphocytes. Obinutuzumab is administered prior to glofitamab in this regimen to reduce the risk of cytokine release syndrome (CRS), a potentially serious immune reaction associated with T-cell engaging therapies. Both drugs are administered intravenously as part of multi-cycle regimens for aggressive lymphomas[1][3][8].
