This combination regimen consists of **glofitamab**, a bispecific CD20xCD3 monoclonal antibody; **obinutuzumab**, a glycoengineered type II anti-CD20 monoclonal antibody; **cyclophosphamide**, an alkylating cytotoxic agent; **doxorubicin**, an anthracycline antibiotic; **vincristine**, a vinca alkaloid; and **prednisone**, a synthetic corticosteroid. Glofitamab leverages a bispecific 2:1 configuration to engage both CD20 on B cells and CD3 on T cells, redirecting T cells to eliminate malignant B cells. Obinutuzumab binds to CD20 on B lymphocytes, leading to direct cell death and enhanced antibody-dependent cellular cytotoxicity. Cyclophosphamide, doxorubicin, and vincristine collectively function as chemotherapeutic agents disrupting cell division via DNA damage, inhibition of topoisomerase II, and mitotic spindle disruption, respectively. Prednisone reduces inflammation and induces apoptosis in lymphoid cells. This regimen is being investigated, primarily for B-cell non-Hodgkin lymphomas, including follicular lymphoma and diffuse large B-cell lymphoma, combining immunotherapy mechanisms with cytotoxic chemotherapy and corticosteroid action[1][3][5][8][9].
