Glofitamab + pirtobrutinib is a combination regimen under investigation for the treatment of relapsed or refractory mantle cell lymphoma (MCL). Glofitamab is a bispecific monoclonal antibody that simultaneously targets CD20 on B-cells and CD3 on T-cells, promoting T-cell-mediated cytotoxicity against malignant B-cells. Pirtobrutinib is a non-covalent, reversible small molecule inhibitor of Bruton's tyrosine kinase (BTK), blocking B-cell receptor (BCR) signaling and impeding proliferation and survival of malignant B-cells, including those with mutations conferring resistance to other BTK inhibitors. The combination aims to provide additive or synergistic antitumor activity and is currently being studied in clinical trials for relapsed/refractory MCL, particularly in patients previously treated with covalent BTK inhibitors[1][2][3][5][7].
