Overview

This is a **six-drug combination regimen** designed primarily for the treatment of B-cell non-Hodgkin lymphoma, especially aggressive and relapsed/refractory forms. The combination includes two monoclonal antibodies (**glofitamab**—a CD20/CD3 bispecific antibody; **rituximab**—an anti-CD20 monoclonal antibody), an **antibody-drug conjugate** (**polatuzumab vedotin**—an anti-CD79b monoclonal antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E), and the classic **CHOP chemotherapy backbone** (**cyclophosphamide**—an alkylating agent, **doxorubicin**—an anthracycline antitumor antibiotic, **prednisone**—a synthetic corticosteroid). Glofitamab acts by redirecting T cells to lymphoma B-cells (via CD3 and CD20), polatuzumab vedotin delivers cytotoxic payload to B-cells via CD79b targeting, rituximab depletes CD20+ B-cells via several immune mechanisms, cyclophosphamide crosslinks DNA, doxorubicin intercalates DNA and generates free radicals, and prednisone induces apoptosis and immune modulation. The combination is being explored to increase efficacy in treatment-resistant lymphomas, leveraging non-overlapping mechanisms of action of all agents.
