Overview

A combination regimen of the **bispecific monoclonal antibody glofitamab** (targeting CD20 x CD3), the anti-CD20 monoclonal antibody **rituximab**, and the classic CHOP chemotherapy backbone (**cyclophosphamide**, **doxorubicin**, **vincristine**, and **prednisone**). - **Glofitamab**: A CD20 x CD3 T-cell–engaging bispecific monoclonal antibody that recruits and activates T cells to kill malignant B cells expressing CD20. - **Rituximab**: A chimeric monoclonal antibody targeting CD20, leading to B-cell depletion primarily via antibody-dependent cellular cytotoxicity, complement-mediated cytotoxicity, and direct apoptosis. - **CHOP**: A standard combination chemotherapy consisting of cyclophosphamide (an alkylating agent), doxorubicin (an anthracycline topoisomerase II inhibitor), vincristine (a vinca alkaloid disrupting microtubule assembly), and prednisone (a corticosteroid modulating inflammation and apoptosis in lymphoid cells). This regimen is under investigation as first-line and relapsed/refractory therapy in **diffuse large B-cell lymphoma** and other aggressive B-cell lymphomas[7].
