Granulocyte-macrophage colony-stimulating factor (GM-CSF) neutralizing antibodies are a class of biologic therapies designed to bind to and inhibit the activity of the cytokine GM-CSF. GM-CSF is a multipotent cytokine that plays a critical role in the production, differentiation, and activation of macrophages and granulocytes, and is specifically involved in the polarization of macrophages toward a pro-inflammatory M1 phenotype. By neutralizing GM-CSF, these antibodies aim to reduce systemic and tissue-specific inflammation. Research presented at AHA 2025 suggests that systemic neutralization of GM-CSF can attenuate neuroinflammation in the stellate ganglion and reduce cardiac sympathetic overactivation in models of chronic heart failure (CHF), potentially restoring cardiac autonomic balance. This class includes several clinical-stage monoclonal antibodies, such as lenzilumab and namilumab, which have been investigated for conditions like rheumatoid arthritis, COVID-19-related cytokine storm, and other inflammatory diseases.
