Overview
Golidocitinib + pomalidomide is an investigational combination therapy being evaluated for the treatment of relapsed or refractory peripheral T-cell lymphoma (R/R PTCL). Golidocitinib is a potent and highly selective small molecule inhibitor of Janus kinase 1 (JAK1), which plays a critical role in the JAK-STAT signaling pathway often dysregulated in T-cell malignancies. Pomalidomide is a third-generation immunomodulatory imide drug (IMiD) that binds to the cereblon E3 ubiquitin ligase complex, leading to the degradation of transcription factors like Ikaros and Aiolos, thereby exerting anti-proliferative and pro-apoptotic effects on tumor cells while stimulating immune responses. The combination aims to leverage the synergistic effects of JAK1 inhibition and immunomodulation to overcome resistance in PTCL.
