Drug intelligence / Profile preview
golidocitinib + gemcitabine + pegaspargase is an investigational combination regimen being evaluated for the treatment of T-cell and NK/T-cell malignancies, particularly advanced extranodal natural killer/T-cell lymphoma (ENKTL). The regimen combines golidocitinib, a highly selective oral Janus kinase 1 (JAK1) inhibitor developed by Dizal, with the chemotherapy agents gemcitabine (a nucleoside analog) and pegaspargase (a PEGylated L-asparaginase). Golidocitinib targets the JAK/STAT pathway, which is frequently hyperactivated in ENKTL and other T-cell lymphomas, thereby suppressing tumor cell proliferation and survival. Gemcitabine inhibits DNA synthesis, while pegaspargase depletes systemic L-asparagine, starving the tumor cells of an essential amino acid. This combination is designed to provide synergistic anti-tumor activity by simultaneously targeting oncogenic signaling, DNA replication, and metabolic pathways in aggressive lymphomas.
