Golidocitinib + pegaspargase + anti-PD-1 monoclonal antibody is a combination therapy regimen under investigation for the first-line treatment of advanced extranodal natural killer/T-cell lymphoma (ENKTL). This triplet regimen combines golidocitinib, a selective Janus kinase 1 (JAK1) inhibitor that blocks the JAK/STAT signaling pathway; pegaspargase, a PEGylated L-asparaginase that depletes systemic L-asparagine to starve lymphoma cells; and an anti-PD-1 monoclonal antibody, an immune checkpoint inhibitor that enhances the body's anti-tumor immune response. The combination is designed to address the aggressive nature of ENKTL by simultaneously targeting oncogenic signaling, metabolic vulnerabilities, and immune evasion mechanisms. Preclinical evidence suggests that JAK1 inhibition can reverse resistance to PD-1/PD-L1 inhibitors, potentially enhancing the efficacy of the immunotherapy component.
