This experimental combination regimen is being investigated for the first-line treatment of advanced or non-nasal extranodal NK/T-cell lymphoma (ENKTL). It integrates **golidocitinib**, a selective Janus kinase 1 (JAK1) inhibitor that targets the STAT3 signaling pathway frequently dysregulated in NK/T-cell malignancies, with the established **P-GemOx** cytotoxic backbone (comprising pegaspargase, gemcitabine, and oxaliplatin) and a **PD-1 inhibitor**. The regimen is designed to achieve synergistic anti-lymphoma activity by simultaneously inhibiting oncogenic signaling, depleting essential asparagine, inducing DNA damage, and reversing immune exhaustion. The combination is currently being evaluated in a Phase 2 multicenter trial led by investigator Wei Xu to determine if the addition of JAK1 inhibition and checkpoint blockade to standard chemotherapy improves complete response rates in treatment-naive patients.
