Overview

GPC3-CD89 is an experimental mRNA-based immunotherapy developed by Myeloid Therapeutics for the treatment of Glypican-3 (GPC3) expressing solid tumors, such as hepatocellular carcinoma. The therapy utilizes lipid nanoparticles (LNPs) to deliver mRNA encoding a chimeric receptor that fuses an anti-GPC3 single-chain variable fragment (scFv) with the alpha chain of the human IgA receptor (CD89). Upon intravenous administration, the LNPs are taken up by circulating and tumor-associated myeloid cells. The resulting fusion protein associates with the endogenous Fc receptor gamma chain (FcRγ) to form a functional chimeric antigen receptor (CAR) on the myeloid cell surface, programming them to recognize and eliminate GPC3-positive cancer cells through phagocytosis and the activation of systemic anti-tumor immunity.
