Overview

Granulocyte colony-stimulating factor (G-CSF), also known as colony-stimulating factor 3 (CSF3), is a glycoprotein cytokine and growth factor that regulates the production, differentiation, and mobilization of neutrophils in the bone marrow. Recombinant human G-CSF (such as filgrastim and lenograstim) is widely used clinically to treat or prevent chemotherapy-induced neutropenia, support hematopoietic stem cell mobilization for transplantation, and manage severe chronic neutropenia. G-CSF acts by binding to the G-CSF receptor (CSF3R) on precursor cells in the bone marrow, activating downstream JAK/STAT, MAPK, and PI3K/Akt signaling pathways to stimulate the survival, proliferation, and maturation of neutrophil precursors. In addition to its hematological indications, G-CSF has been investigated by developers such as Beijing 302 Hospital (now the Fifth Medical Center of the PLA General Hospital) for its potential to promote hepatic regeneration and improve survival in patients with acute-on-chronic liver failure (ACLF).
