GSK5926371 is an investigational B-cell depleting T-cell engager (TCE) developed by GSK for the treatment of autoimmune rheumatic diseases (ARD). The molecule is designed to simultaneously bind to CD19 and CD20 on B-lymphocytes and CD3 on T-lymphocytes. This dual-targeting approach aims to achieve more comprehensive B-cell depletion than therapies targeting only one marker (like rituximab), potentially eliminating pathogenic B-cell populations that contribute to chronic inflammation. It is currently being evaluated in the Phase 1 ELEVATE-1 clinical trial for several B-cell driven conditions, including systemic lupus erythematosus, rheumatoid arthritis, idiopathic inflammatory myopathies, and Sjogren's disease.
