GT19077 is a small molecule inhibitor designed to disrupt the protein-protein interaction (PPI) between the oncogenic transcription factor c-Myc and its obligate partner Max. Developed by Kintor Pharmaceutical in collaboration with Peking University, the compound targets c-Myc-driven malignancies, including B-cell lymphomas with IGH/Myc translocations and small cell lung cancer (SCLC). By preventing the formation of the c-Myc/Max heterodimer, GT19077 inhibits the transcriptional activity of c-Myc, leading to G2/M cell cycle arrest and the down-regulation of immune evasion markers such as CD47 and PD-L1. Preclinical studies have demonstrated that GT19077 selectively inhibits the proliferation of Myc-dependent tumor cells while sparing normal hematopoietic progenitor cells, suggesting a favorable therapeutic index. Additionally, the compound has been shown to promote the degradation of c-Myc protein in cellular models.
