A combination therapy consisting of guadecitabine (a hypomethylating agent), ipilimumab (an anti-CTLA-4 monoclonal antibody), and nivolumab (an anti-PD-1 monoclonal antibody). This triple combination is designed to enhance anti-tumor immune responses through complementary mechanisms: guadecitabine promotes DNA demethylation and activates innate immunity pathways, while ipilimumab and nivolumab block different immune checkpoints to promote T-cell function and tumor killing. The combination is being investigated primarily for patients with melanoma and non-small cell lung cancer who have developed resistance to prior anti-PD-1/PD-L1 therapy.
