Overview

h20.19 CAR T cells is an investigational, autologous, bispecific chimeric antigen receptor (CAR) T-cell therapy developed by the Medical College of Wisconsin. This cell-based immunotherapy is engineered using a lentiviral vector to express a tandem CAR targeting two distinct B-cell antigens: CD19 and CD20. The construct incorporates fully human binding domains along with 4-1BB (CD137) costimulatory and CD3ζ signaling domains. By targeting two antigens simultaneously, h20.19 is designed to improve clinical durability and mitigate the risk of disease relapse caused by antigen escape or downregulation, a common limitation of single-target CAR-T therapies. The treatment is primarily being evaluated for patients with relapsed or refractory B-cell malignancies, including B-cell non-Hodgkin lymphoma (NHL) and chronic lymphocytic leukemia (CLL). Manufacturing is conducted using the CliniMACS Prodigy automated system at the Medical College of Wisconsin's Cell Therapy Laboratory.
