hAB08-MTI1 is an antibody-drug conjugate (ADC) developed by Pfizer, consisting of a humanized monoclonal antibody (hAB08) targeting the interleukin-13 receptor alpha 2 (IL-13Rα2) conjugated to a novel microtubule inhibitor (MTI1). IL-13Rα2 is a transmembrane protein highly expressed in various cancers, including glioblastoma, ovarian, and pancreatic cancers, and is often upregulated in chemotherapy-resistant tumor cells. Upon binding to IL-13Rα2, the ADC is internalized, allowing the microtubule inhibitor payload to exert its cytotoxic effect. Preclinical studies have demonstrated potent antitumor activity in IL-13Rα2-positive models such as prostate cancer (PC3MM2) and melanoma (A375).
