hAB08-MTI2 is an antibody-drug conjugate (ADC) developed by Pfizer for the treatment of various cancers, including glioblastoma, ovarian, and pancreatic cancers. It consists of a humanized monoclonal antibody, hAB08, which binds with high affinity to the interleukin-13 receptor alpha 2 (IL-13Rα2), a protein frequently overexpressed in these malignancies and associated with chemotherapy resistance. The antibody is conjugated to MTI2, a novel microtubule inhibitor payload. Upon binding to IL-13Rα2, the ADC is internalized into the tumor cell, where the payload is released to disrupt microtubule dynamics, leading to cell cycle arrest and apoptosis. Preclinical studies have demonstrated significant antitumor efficacy in models of prostate cancer and melanoma.
