Haitrombopag ethanolamine is a small-molecule, non-peptide thrombopoietin receptor (TPO-R) agonist developed by Jiangsu Hengrui Pharmaceuticals. It acts as an allosteric agonist by binding to the transmembrane domain of the TPO receptor (also known as MPL), which triggers intracellular signaling pathways, including the JAK/STAT and MAPK pathways. This stimulation promotes the proliferation and differentiation of megakaryocytes from bone marrow progenitor cells, leading to increased platelet production. Haitrombopag was approved by the China National Medical Products Administration (NMPA) in 2021 for the treatment of chronic immune thrombocytopenia (ITP) in patients who have had an insufficient response to other treatments, and it is also indicated for severe aplastic anemia (SAA).
