HBW-012462 is an orally bioavailable small molecule pan-KRAS inhibitor developed by Hyperway Pharmaceutical. It is designed to target a broad spectrum of KRAS mutations by simultaneously inhibiting both the active (ON) and inactive (OFF) states of the KRAS protein. This dual-state inhibition mechanism is intended to provide broader clinical utility compared to mutation-specific inhibitors, which often only target the inactive state of specific variants like G12C. Preclinical data indicate that HBW-012462 possesses high potency, selectivity, and advantageous tissue targeting properties. The drug received clinical trial approval from China's National Medical Products Administration (NMPA) in October 2025 and is currently being evaluated in Phase 1 clinical trials for the treatment of various KRAS-driven solid tumors.
