HDM2005 + rituximab + lenalidomide is a combination therapy regimen currently under clinical investigation for the treatment of relapsed or refractory mantle cell lymphoma (MCL). The regimen consists of **HDM2005**, a selective and irreversible Bruton's tyrosine kinase (BTK) inhibitor developed by Huadong Medicine, in combination with the anti-CD20 monoclonal antibody **rituximab** and the immunomodulatory drug (IMiD) **lenalidomide**. This triplet combination is designed to provide synergistic anti-tumor activity by simultaneously blocking B-cell receptor signaling (via BTK inhibition), inducing antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity (via CD20 targeting), and modulating the immune microenvironment while inducing the degradation of specific transcription factors (via cereblon binding). The combination is being evaluated in a Phase Ib/III trial (NCT06468241) to determine if it offers superior clinical benefit compared to standard rituximab-based regimens in patients who have previously failed treatment with a BTK inhibitor.
