Overview

Hexavalent epratuzumab (Hex-h22) is a multivalent antibody construct engineered using the proprietary Dock-and-Lock (DNL) platform. It is composed of six humanized Fab fragments derived from epratuzumab, a monoclonal antibody that targets the CD22 antigen expressed on B-lymphocytes. By increasing the valency from two (in the standard bivalent antibody) to six, Hex-h22 exhibits significantly higher avidity for CD22. This enhanced binding strength facilitates more potent induction of CD22 internalization and more effective modulation of B-cell receptor (BCR) signaling compared to the parent antibody. These mechanisms result in superior growth inhibition and apoptosis in B-cell malignancies. Developed by Immunomedics (now part of Gilead Sciences), Hex-h22 has been evaluated in preclinical and early-stage research for the treatment of B-cell hematologic cancers, such as non-Hodgkin lymphoma and chronic lymphocytic leukemia.
