Hexavalent hA20 (Hex-hA20) is a novel hexavalent anti-CD20 antibody biologic engineered using the proprietary Dock-and-Lock (DNL) platform technology. The construct is composed of a humanized anti-CD20 IgG (veltuzumab, also known as hA20) serving as the foundation, to which four additional anti-CD20 hA20 Fab fragments are attached, resulting in a single molecule with six antigen-binding sites. This high-valency design is intended to enhance the therapeutic index by inducing potent antibody-dependent cell-mediated cytotoxicity (ADCC), direct antiproliferative effects, and homotypical adhesion in CD20-positive cells. Developed by IBC Pharmaceuticals, Hex-hA20 has shown significant antitumor efficacy in preclinical models of B-cell lymphoma and non-Hodgkin lymphoma, demonstrating subnanomolar potency in vitro.
