Overview

HF0299 is an investigational, orally bioavailable small molecule inhibitor that selectively targets Janus kinase 1 (JAK1). Developed by Haisen Pharmaceutical, it is designed to modulate the JAK-STAT signaling pathway, which is a central driver of inflammation in various immune-mediated diseases. By specifically inhibiting the JAK1 isoform, HF0299 blocks the downstream signaling of key pro-inflammatory cytokines, including interleukin-4 (IL-4), IL-13, and IL-31, which are critical to the pathogenesis of moderate-to-severe atopic dermatitis. The drug's selectivity for JAK1 over other family members, particularly JAK2, is intended to minimize off-target effects such as anemia and myelosuppression. HF0299 is currently undergoing clinical evaluation to establish its safety and efficacy as a systemic treatment for moderate-to-severe atopic dermatitis.
