HinTCAR2019 is a hinge-optimized, tandem bispecific chimeric antigen receptor (CAR) T-cell therapy targeting both CD19 and CD20 antigens. Developed to address antigen escape in patients with relapsed or refractory B-cell lymphoma (r/r BCL), the construct features a minimal hinge optimization—specifically a short "AS" motif between the scFv and hinge domain—which enhances CAR surface expression and cytotoxic potency compared to earlier dual CAR constructs. Clinical data from a Phase Ia/Ib study (NCT05388695) indicates a favorable safety profile and durable efficacy, particularly when used in a sequential strategy followed by autologous stem cell transplantation (ASCT) consolidation.
