Overview

HLA-G inhibitors are a class of therapeutic agents designed to block the activity of Human Leukocyte Antigen G (HLA-G), a non-classical MHC class I molecule. HLA-G is normally expressed in the placenta to provide fetal-maternal tolerance but is frequently neo-expressed by various tumors to evade the immune system. By binding to inhibitory receptors such as ILT2 and ILT4 on immune cells (NK cells, T cells, and myeloid cells), HLA-G suppresses anti-tumor immune responses. Inhibition of HLA-G, often through monoclonal antibodies or gene-silencing techniques, aims to restore immune surveillance and is being investigated for the treatment of solid tumors and brain metastases. Research indicates that HLA-G may also interact with the SPAG9-STAT3 signaling axis to promote metastatic progression, making its inhibition a potential strategy to prevent the formation of mature brain lesions.
