HRS-717 + HRS-4642 + SHR-1316 (also referred to as HRS-7172 + HRS-4642 + SHR-1316) is an investigational combination regimen developed by Jiangsu Hengrui Pharmaceuticals for the treatment of advanced solid tumors, particularly advanced pancreatic cancer harboring RAS mutations or amplifications. The combination consists of three active components: HRS-7172, an orally active pan-RAS inhibitor; HRS-4642, an intravenously administered liposomal formulation of a selective KRAS G12D inhibitor; and SHR-1316 (adebrelimab), a humanized IgG4 monoclonal antibody targeting the immune checkpoint PD-L1. By simultaneously inhibiting oncogenic RAS/KRAS signaling and blocking the PD-1/PD-L1 immune checkpoint pathway, this combination is designed to overcome the immunosuppressive tumor microenvironment and enhance anti-tumor efficacy in RAS-mutated cancers.
