Overview

**Hu1928-Hu20BB CAR T cells** are an autologous bicistronic CD19/CD20-directed CAR-T-cell therapy developed by the National Cancer Institute for relapsed or refractory B-cell malignancies. Patient T cells are gamma-retrovirally transduced with an MSGV1 vector encoding two fully human CARs: Hu19-CD828Z, an anti-CD19 CAR containing CD28 and CD3ζ signaling domains, and Hu20-CD8BBZ, an anti-CD20 CAR containing 4-1BB and CD3ζ signaling domains. Following intravenous infusion, the engineered T cells recognize and eliminate CD19-positive and/or CD20-positive malignant B cells, providing a strategy to reduce antigen-escape failure after CD19-directed CAR-T therapy. The original construct underwent sequence optimization to reduce retroviral recombination; Hu1928-Hu20BB-standard and Hu1928-Hu20BB-long are distinct optimized construct variants and are not treated here as synonyms of the exact unqualified Hu1928-Hu20BB CAR-T-cell product.
