HXN-2001 is a first-in-class biparatopic dual-payload antibody-drug conjugate (ADC) developed by Earendil Labs (Helixon) for the treatment of colorectal cancer. It specifically targets Cadherin-17 (CDH17), a calcium-dependent cell adhesion protein that is highly expressed in gastrointestinal malignancies but has limited expression in normal tissues. The drug's biparatopic design allows it to bind to two non-overlapping epitopes on the CDH17 extracellular domain, which typically enhances receptor internalization and lysosomal trafficking compared to monospecific antibodies. Furthermore, HXN-2001 is engineered with a dual-payload system, delivering two distinct cytotoxic agents to the tumor cell to maximize therapeutic efficacy and potentially overcome drug resistance. It is currently being evaluated in Phase 1 clinical trials.
