HXN-2013 is a next-generation biparatopic antibody-drug conjugate (ADC) targeting B7-H3 (CD276), currently under development by Earendil Labs (formerly Helixon) for the treatment of small-cell lung cancer (SCLC). B7-H3 is an immune checkpoint molecule of the B7 family that is frequently overexpressed in SCLC and various other solid tumors, where it is associated with tumor progression and immune evasion. HXN-2013 utilizes a biparatopic antibody design that recognizes two distinct, non-overlapping epitopes on the B7-H3 protein. This dual-binding approach is engineered to promote superior receptor clustering and rapid internalization of the ADC-target complex into lysosomal compartments, thereby maximizing the intracellular release of its cytotoxic payload. By specifically delivering a potent toxin to B7-H3-expressing malignant cells, HXN-2013 aims to provide a targeted therapeutic option with an improved safety profile and enhanced anti-tumor activity compared to conventional chemotherapy or monoparatopic ADCs.
