This investigational combination regimen consists of hypofractionated radiotherapy, granulocyte-macrophage colony-stimulating factor (GM-CSF), pomalidomide, and glofitamab. It is currently being evaluated in a Phase 2 clinical trial for the treatment of newly diagnosed primary central nervous system diffuse large B-cell lymphoma (PCNSL DLBCL). The regimen aims to combine the immunogenic cell death induced by radiation with the immunostimulatory effects of GM-CSF, the immunomodulatory properties of pomalidomide (a cereblon E3 ligase modulator), and the T-cell redirecting activity of glofitamab (a CD20xCD3 bispecific antibody). This multi-modal approach is designed to leverage synergistic immune activation to overcome the aggressive nature and immune-privileged environment of CNS lymphomas.
