Overview

IA-Dimer is a dimeric, non-peptide antagonist of the integrin αvβ3, designed to inhibit tumor angiogenesis and growth. It is a multivalent version of the monomeric antagonist 4-[2-(3,4,5,6-tetrahydropyrimidine-2-ylamino)ethyloxy]benzoyl-2-(S)-aminoethylsulfonyl-amino-β-alanine. By targeting the αvβ3 integrin, which is upregulated on angiogenic endothelial cells, IA-Dimer aims to disrupt tumor blood vessel formation. In preclinical evaluations using B16F10 melanoma tumor-bearing mice, IA-Dimer demonstrated the ability to slow tumor growth, although it showed lower efficacy and higher toxicity compared to its trimeric counterpart, IA-Trimer. The research was conducted by investigators at the Methodist Hospital Research Institute and Weill Medical College of Cornell University.
