IBI3020 is a first-in-class dual-payload antibody-drug conjugate (ADC) developed by Innovent Biologics using their proprietary DuetTx® platform. It targets CEACAM5 (carcinoembryonic antigen-related cell adhesion molecule 5, also known as CEA), a cell-surface glycoprotein that is overexpressed in various solid tumors, including colorectal, non-squamous non-small cell lung, gastric, and pancreatic cancers, while maintaining limited expression in healthy tissues. IBI3020 is designed to selectively bind to CEACAM5-expressing tumor cells, followed by internalization and lysosomal degradation, which releases two distinct types of clinically validated cytotoxic payloads. This dual-payload approach is intended to enhance antitumor efficacy and provide a robust bystander killing effect. As of April 2025, IBI3020 became the first dual-payload ADC globally to complete first-in-human dosing and is currently undergoing Phase 1 clinical evaluation in the United States and China.
