iDAb RAS-UBOX is an experimental pan-RAS macromolecule degrader (macrodrug) designed to induce the proteolysis of all three RAS isoforms: KRAS, NRAS, and HRAS. It is a fusion protein consisting of a pan-RAS-binding intracellular single domain antibody (iDAb RAS) and the UBOX domain of the carboxyl terminus of Hsc70-interacting protein (CHIP) E3 ligase. By recruiting the ubiquitin-proteasome system, the construct facilitates the degradation of endogenous RAS proteins. Research indicates that while it effectively inhibits the proliferation of various cancer cell lines regardless of the specific RAS mutation, it also affects non-transformed cells expressing wild-type RAS, distinguishing its profile from isoform-specific degraders. It was developed by researchers at the University of Oxford as a tool to study RAS biology and as a potential therapeutic strategy for RAS-driven cancers.
