Overview

idasanutlin + obinutuzumab is a combination of two anticancer agents: - **Idasanutlin** is a selective small molecule antagonist of MDM2, a negative regulator of the tumor suppressor protein p53. By inhibiting the interaction between MDM2 and p53, idasanutlin activates the p53 pathway, leading to increased cancer cell apoptosis, especially in p53 wild-type tumors. - **Obinutuzumab** is a glycoengineered, humanized, type II IgG1 monoclonal antibody targeting the CD20 antigen on B cells. It induces B cell lysis primarily through enhanced antibody-dependent cellular cytotoxicity (ADCC), direct non-apoptotic cell death, and, to a lesser extent, complement-dependent cytotoxicity (CDC). This combination has demonstrated preclinical and early clinical antitumor activity in B-cell malignancies such as follicular lymphoma, especially in p53 wild-type tumors. The combination leverages complementary mechanisms: idasanutlin drives p53-mediated apoptosis, and obinutuzumab maximizes B-cell depletion via immune-mediated and direct cytotoxic effects[1][3][4][5][6].
