Overview

**Idasanutlin + rituximab** is an experimental drug combination consisting of idasanutlin, a selective small molecule MDM2 antagonist, and rituximab, a monoclonal anti-CD20 antibody. Idasanutlin targets the p53 pathway by inhibiting the interaction between MDM2 and p53, leading to increased p53 activity, cell cycle arrest, and apoptosis in tumor cells harboring wild-type p53. Rituximab binds to CD20 on B cells, mediating B-cell depletion primarily through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity. The combination is being developed for B-cell lymphoid malignancies, specifically follicular lymphoma (FL) and diffuse large B-cell lymphoma (DLBCL). Preclinical and early clinical data support synergistic anti-tumor activity, with idasanutlin enhancing rituximab-induced apoptosis without negatively affecting rituximab's immune effector functions[1][2][5][6].
