Overview

Idelalisib + rituximab is a combination therapy used primarily for the treatment of relapsed chronic lymphocytic leukemia (CLL). Idelalisib is an oral small molecule inhibitor that selectively targets the delta isoform of phosphoinositide 3-kinase (PI3Kδ), a key enzyme in B-cell receptor signaling pathways. By inhibiting PI3Kδ, idelalisib disrupts malignant B-cell proliferation and survival. Rituximab is a monoclonal antibody that binds to CD20 on the surface of B-cells, marking them for destruction by the immune system. This combination exploits both targeted inhibition of intracellular signaling and immune-mediated cell killing to control CLL progression[1][2][5][6].
