Overview

Ifidancitinib is a small molecule Janus kinase (JAK) inhibitor that selectively targets JAK1 and JAK3. Developed by Aclaris Therapeutics in collaboration with Rigel Pharmaceuticals, the drug disrupts the signaling of cytokines that utilize the common gamma chain (γc) receptor, a pathway critical to the pathogenesis of various autoimmune and inflammatory dermatological conditions. Ifidancitinib was primarily developed as a topical formulation (ATI-502) for the treatment of alopecia areata, vitiligo, and atopic dermatitis. It also serves as the active moiety for the oral prodrug fosifidancitinib (ATI-501). Clinical development reached Phase 2, where it demonstrated efficacy in hair regrowth; however, Aclaris subsequently sought a strategic partner for the program to refocus on its core pipeline assets.
