Drug intelligence / Profile preview
An IL-10RA blocker is a therapeutic agent (such as a monoclonal antibody or peptide) designed to bind to the Interleukin-10 receptor subunit alpha (IL-10RA, also known as CD210) and inhibit its interaction with the immunosuppressive cytokine Interleukin-10 (IL-10). IL-10 signaling normally limits inflammatory responses, but its prolonged or elevated systemic production can lead to pathological immunosuppression in chronic infections and various cancers. By blocking the IL-10/IL-10RA pathway, these agents prevent downstream JAK1/TYK2/STAT3 activation, thereby reversing macrophage-mediated immunosuppression, promoting protective proinflammatory macrophage phenotypes (such as CD163+CD206+ macrophages), and enhancing host-protective T-cell and immune effector functions against viral infections (such as Severe Fever with Thrombocytopenia Syndrome [SFTS] and COVID-19) and tumors.
