IL2-NP is a nanoparticle-formulated interleukin-2 (IL-2) therapeutic candidate developed by PDX Pharmaceuticals using its proprietary Pdx-NP™ platform. The 160-nm nanoconstruct is designed to improve the therapeutic index of IL-2 by enhancing tumor accumulation and extending circulation half-life, addressing the limitations of free IL-2 such as rapid clearance and systemic toxicity. IL2-NP acts as an agonist for the interleukin-2 receptor, promoting the expansion and activation of tumor-infiltrating lymphocytes (TILs), specifically CD8+ T cells, while potentially mitigating the counterproductive activation of regulatory T cells (Tregs). Preclinical studies in melanoma and colon cancer models have demonstrated that IL2-NP induces robust anti-tumor immune responses both as a monotherapy and in combination with immune checkpoint inhibitors, showing significant growth reduction and improved survival in mouse models.
