Drug intelligence / Profile preview
IMM0306S is an investigational subcutaneous formulation of amulirafusp alfa (IMM0306), a first-in-class bispecific CD20×CD47 antibody fusion protein developed by ImmuneOnco Biopharmaceuticals. Built on the proprietary 'mAb-Trap' platform, the molecule consists of a CD20-targeting monoclonal antibody fused to the CD47-binding domain of SIRPα. This dual-targeting mechanism simultaneously binds CD20 on B cells and CD47, blocking the CD47-SIRPα 'don't eat me' inhibitory signal while promoting B-cell depletion via Fcγ receptor-mediated phagocytosis and NK cell activation. Unlike some other CD47-targeting therapies, amulirafusp alfa is designed to avoid binding to human red blood cells, reducing the risk of treatment-induced anemia. IMM0306S is being developed for B-cell-mediated autoimmune diseases, with clinical evaluation currently underway in a Phase Ib trial for systemic lupus erythematosus (SLE).
