Overview

IMPT-514 is an investigational, autologous, bispecific chimeric antigen receptor (CAR) T-cell therapy designed to target both CD19 and CD20 antigens. It utilizes a potent bispecific CAR construct with a 4-1BB costimulatory domain and is administered as a single intravenous infusion following lymphodepleting chemotherapy. The therapy aims to deplete autoreactive B cells and pathogenic CD20-expressing T-cells implicated in autoimmune diseases such as systemic lupus erythematosus (SLE), lupus nephritis (LN), and multiple sclerosis (MS). By targeting both CD19 and CD20, IMPT-514 can eliminate B cells at various stages of maturation as well as certain pro-inflammatory T-cell subsets, potentially resetting the immune system with durable remission. Developed by ImmPACT Bio, it is the first dual-targeting CAR-T cell therapy for lupus indications and the only such therapy currently in development for MS[1][2][3][4][5][6][7].
