Overview

This is an investigational **three-drug combination therapy** consisting of **INCB047986 (a selective Janus kinase 1, or JAK1, inhibitor)**, **gemcitabine (an antimetabolite chemotherapeutic agent that inhibits DNA synthesis)**, and **nab-paclitaxel (a nanoparticle albumin-bound formulation of the microtubule inhibitor paclitaxel)**. INCB047986 is designed to inhibit JAK1-mediated signaling, thereby limiting tumor cell proliferation and modulating the tumor microenvironment; gemcitabine blocks DNA synthesis, leading to cell death; nab-paclitaxel disrupts microtubule function to prevent cell division and facilitate apoptosis. This combination has been or is being studied as a potential treatment in cancer, including pancreatic cancer, with the goal of combining direct cytotoxic effects (from gemcitabine and nab-paclitaxel) with the immunomodulatory and antiproliferative effects of JAK1 inhibition (from INCB047986).
